Once ADC drugs enter the bloodstream, they attach to target antigen receptors on the tumor cell surface. The resulting ADC-antigen complex is internalized and broken down by lysosomes, releasing the payload to induce tumor cell death. As a critical component of ADCs, the payload's activity and physicochemical properties directly influence the anti-tumor effectiveness of these drugs. An ideal payload should exhibit high cytotoxicity, low immunogenicity, strong stability, modifiable functional groups, a bystander killing effect, suitable water solubility, and intracellular targeting. Common types of payloads include tubulin inhibitors and DNA-damaging agents.
