Crosslinkers primarily serve to link antibodies with toxins. In Antibody-Drug Conjugates (ADCs), the linker connecting cytotoxic drugs to monoclonal antibodies is a critical component. One of the primary challenges in developing effective ADCs lies in selecting an appropriate crosslinker. This crosslinker must ensure the cytotoxic drug is released in its active form within or near the target cell. It plays a fundamental role in the efficient delivery of cytotoxic drugs via ADCs and significantly influences the toxicity of ADC products. Premature drug release into the bloodstream can result in systemic toxicity and a reduced therapeutic index. The selection of a linker typically depends on factors such as the target, the drug, the internalization and breakdown of the antibody-antigen complex, as well as the characteristics of the drug being used.
