Once ADC drugs enter the bloodstream, they attach to target antigen receptors on the surface of tumor cells. The resulting ADC-antigen complex is internalized and broken down by lysosomes, releasing the payload to induce tumor cell death. As a critical component of ADCs, the payload's activity and physicochemical properties have a direct impact on the anti-tumor effectiveness of these drugs. An ideal payload should possess high cytotoxicity, low immunogenicity, strong stability, modifiable functional groups, a bystander killing effect, suitable water solubility, and an intracellular target. Commonly used payloads include tubulin inhibitors and DNA-damaging agents.
