Enhancer of zeste homolog 2 (EZH2), a well-known epigenetic regulator, serves as the catalytic subunit of the Polycomb Repressive Complex 2 (PRC2), which silences target genes by tri-methylating lysine 27 on histone H3 (H3K27me3). EZH2 plays a pivotal role in both development and the maintenance of adult tissue homeostasis, while also being strongly linked to various diseases. Research has shown that EZH2 is overexpressed in numerous solid tumors and hematological malignancies, contributing to tumor growth, invasion, metastasis, and poor clinical outcomes. As a result, EZH2 represents a promising pharmacological target for cancer treatment.
