Poly (ADP-ribose) polymerase (PARP) refers to a family of enzymes crucial for DNA repair, as they facilitate the addition of ADP-ribose units to proteins, a process vital for addressing DNA damage. Among them, PARP1 and PARP2 are the most prominent, playing significant roles in repairing single-strand DNA breaks and preserving genomic stability. Due to their involvement in DNA repair, PARPs have emerged as promising targets for cancer therapy, especially in tumors with deficiencies in other DNA repair pathways, such as those with BRCA1/2 mutations. By inhibiting PARP, synthetic lethality can be induced, effectively targeting cancer cells with compromised DNA repair capabilities.
