Protein arginine methyltransferase 5 (PRMT5) is a key type II arginine methyltransferase responsible for catalyzing the formation of symmetric dimethylarginine in various nuclear and cytoplasmic proteins. PRMT5 is crucial for regulating essential biological processes such as transcription, cell cycle progression, RNA splicing, and DNA repair. Recent studies have increasingly highlighted PRMT5's role as a tumor-promoting factor in multiple cancers. It enhances the carcinogenicity of numerous solid tumors, including those of the colon, breast, prostate, lung, liver, bone, ovary, stomach, and pancreas, often associated with poor clinical outcomes. In recent years, innovative inhibitors targeting the PRMT5/MTA complex have been developed as potential treatments for MTAP-deleted cancers.
