Interleukin-1 Receptor-Associated Kinase 4 (IRAK4) is one of four kinases in the IRAK family, which also includes IRAK1, IRAK2, and IRAK-M. As a serine/threonine kinase, IRAK4 plays a pivotal role in the signaling pathways of Toll-Like Receptors (TLR) and Interleukin-1 Receptors (IL-1R). Following TLR activation, IRAK4 is recruited by the adaptor protein Myeloid Differentiation Primary Response Gene 88 (MyD88), leading to the activation of nuclear factor kappa B (NF-κB) and type I interferon (IFN) pathways. Dysregulated IRAK4 activity has been implicated in various autoimmune diseases, including rheumatoid arthritis (RA) and systemic lupus erythematosus (SLE), as well as conditions like lymphocytic leukemia, lymphoma, and fibrotic disorders. As a result, IRAK4 inhibitors hold potential as therapeutic agents for treating inflammatory diseases and certain cancers.
