The NLRP3 inflammasome, a multimeric protein complex, consists of three structural domains: the leucine-rich repeat (LRR) domain, the nucleotide-binding and oligomerization (NACHT) domain, and the pyrin domain (PYD). As a distinctive pattern recognition receptor (PRR), NLRP3 can be broadly activated by bacterial, viral, and fungal pathogens, as well as environmental factors and host-derived signals. Dysregulated activation of the NLRP3 inflammasome has been linked to the development of numerous human diseases, contributing to both acute and chronic inflammation-associated conditions, such as arthritis, atherosclerosis, diabetes, multiple sclerosis, Alzheimer's disease, and Parkinson's disease. Consequently, targeting the NLRP3 inflammasome with specific inhibitors holds potential as a therapeutic strategy for these conditions.
