Certain enzymes within intracellular lysosomes and the tumor microenvironment have the ability to selectively cleave specific linkers. For instance, cathepsins in lysosomes can hydrolyze peptide bonds, including dipeptide or tetrapeptide linkers. Additionally, glycosidases can break down β-glucuronide and β-galactosidase, phosphatases can hydrolyze pyrophosphate, and sulfatases are capable of hydrolyzing sulfates.
